To get the sign on the River level, wait until you come to the place where the Vileplume is sleeping. Play your pokeflute and it will wake up. All those fumes coming from it, which were blocking the sign, will have disappeared. Look up and you will see that the tree that was blocked by the fumes is now visable. It is a Cubone. That's the sign. Take a picture of it and then show Prof. Oak.

use the pokeflute on vileplume and youll see a cubone tree
