When you get the "Pick Nails" in the Egg Quaters level and have that level completed, go back to the "Dry Lagoon" level using level select on the main menu.

Using the pick nails, have Rouge dig into the painting just above the first ledge on the east wall. You land in a small room where the Mystic Melody waits.
