There are many unlockables, so I suggest you check out an FAQ:

Try that one; scroll down to the section called "SECRETS" which tells you what you get for a 1st place trophy on each of the races, at each level.

You get them by winning races.

You get Toad and Toadette by winning Special Cup on 100cc. You get Petey Piranha and King Boo by winning Star Cup on Mirror Mode.
