You don't need to have the Alley-Oop ability unlocked. Once your House meter is full and the crowd is chanting 'House Juice' or something to that effect, run towards the basket holding the L Trigger and R Trigger and press 'Y' button. If timed right, and the defender doesn't get in the way, you should 'Bring Down the House'. I'm still amazed they didn't cover this in the game manual. Good luck!
